Impact of free heme on the cellular functions of a mouse macrophage line and a mouse B lymphocyte line UQAM - Montréal

 

The aim of this project will be to study the effects of free heme (haemin) on immune cells (macrophages, T lymphocytes) and on cells involved in bone remodelling (osteoclasts and osteoblasts). Despite the existence of proteins capable of binding and transiently blocking the reactivity of haem (haemopexin, albumin) with cellular structures, as well as the induction of the enzyme haem oxygenase 1 (HO-1), responsible for the degradation of haem, haemolytic conditions can lead to their saturation. During this project, various techniques will be used such as immunoblot (WB), flow cytofluorometry, measurement of metabolic activity, RT-PCR and ELISA.
